Marion SCHILLING"},"content":{"rendered":"<h2><span class=\"fontstyle0\">From forests to vineyards: adaptation of wood-degrading fungi<\/span><\/h2>\n<p>Defended on December 16, 2022, at the Faculty of Sciences and Technology,\u00a0 Vandoeuvre-L\u00e8s-Nancy<\/p>\n<p>Thesis supervosors: Dr. Eric GELHAYE Lorraine University, Vandoeuvre-L\u00e8s-Nancy and <strong>Dr. Sibylle FARINE<\/strong>, University Haute Alsace<\/p>\n<table class=\"NormalTable\" width=\"5\">\n<tbody>\n<tr>\n<td width=\"200\"><\/td>\n<\/tr>\n<\/tbody>\n<\/table>\n<p><span class=\"fontstyle0\">Abstract<br \/>\n<\/span><span class=\"fontstyle1\">Esca is one of the most devastating of the grapevine trunk diseases, that affect vineyards around the world.<br \/>\nOne of its main symptoms is white rot, also called <\/span><span class=\"fontstyle3\">amadou<\/span><span class=\"fontstyle1\">. In white rot, <\/span><em><span class=\"fontstyle3\">Fomitiporia mediterranea <\/span><\/em><span class=\"fontstyle1\">(M.<br \/>\nFischer) is the major species identified in Europe. White-rot fungi have evolved with their substrates and<br \/>\nmay have developed specifically adapted degradation and detoxification mechanisms.<br \/>\nThe main objective of this thesis is to understand how <\/span><em><span class=\"fontstyle3\">F. mediterranea <\/span><\/em><span class=\"fontstyle1\">is adapted to its substrate, grapevine<br \/>\nwood. The behavior of <\/span><em><span class=\"fontstyle3\">F. mediterranea <\/span><\/em><span class=\"fontstyle1\">during wood degradation on different wood species was monitored.<br \/>\nCarbon mineralization rate, wood mass loss, chemical composition in structural compounds and extractives,<br \/>\nfungal biomass production and protein secretion (secretome) upon cultures on sawdust were measured. In<br \/>\na second step, the colonization of <\/span><em><span class=\"fontstyle3\">F. mediterranea <\/span><\/em><span class=\"fontstyle1\">on wood blocks was followed. In addition to the<br \/>\nphysiological approach, a non-targeted metabolomic approach, with molecular networking, was used to<br \/>\ndescribe fungal and wood metabolomes during colonization.<br \/>\n<\/span><span class=\"fontstyle3\"><em>F. mediterranea<\/em> <\/span><span class=\"fontstyle1\">is able to degrade several wood species, including grapevine cv. \u2018Gewurztraminer\u2019 and<br \/>\n\u2018Riesling\u2019, beech and oak, in sawdust or blocks, using a simultaneous degradation pattern. Parallely, the<br \/>\nfungus regulates its secretome in relation with its substrate: a larger number of oxidoreductases were<br \/>\nsecreted on grapevine sawdust. This could be correlated with the observed degradation of grapevine lignin<br \/>\nand specialized metabolites, mainly stilbenes.
